CMAP x GTEx - Sex-Biased Drug Response Explorer

How do male and female cells differ when exposed to the same drug? This explores per-lineage GSEA results (CMAP/LINCS 2020 vs GTEx v8) from Project 59.

What is this?

Drug trials usually report a single average effect. But male and female cells often respond to the same drug differently. This app explores how a drug's sex-specific effect on gene expression lines up with a tissue's natural sex biology, using per-lineage GSEA on CMAP/LINCS 2020 drug signatures vs GTEx v8 sex-biased genes.

How to use it

  1. Use the Select a drug dropdown below. Pick All to see every hit, or type a drug name (e.g. tozasertib) to focus on one.
  2. Read the table: each row is a drug-tissue pair where the drug significantly overlaps that tissue's sex-biased genes (FDR < 0.05).
  3. Read the bar chart: it shows the NES (enrichment score) per hit.

How to read the numbers

  • Green bar (NES above 0) -> effect leans male-biased
  • Red bar (NES below 0) -> effect leans female-biased
  • FDR = reliability; smaller is stronger (all shown are < 0.05)

Categories

  • A - drug hits sex-biased genes in its OWN tissue -> real signal (tozasertib, lapatinib, vorinostat)
  • B - drug hits sex-biased genes in a DIFFERENT tissue -> off-target candidate (23 drugs)
  • C - no significant sex-specific effect (not shown here)
